The vitamin D plays multifold roles in the regulation of metabolism. There are two forms: Firstly, at the plasma membrane level, secondly, at the gene transcription level. The vitamin D receptor is popularly known as the VDR.

The receptors for the steroid and thyroid hormones are located inside the target this occurs in cells, in the cytoplasm or the nucleus and function ligand-dependent transcription factors. The key factors in these are the amino acid terminus, DNA binding domain and the carboxy-terminus or the ligand binding -domain.

The VDR has a domain structure homologous to all its receptors. It has four hundred and twenty -seven (427) amino acids. It has a short N-ten, minal activation function-1 (AF-1); this also has a DNA Binding domain, a flexible hinge region. It has an HSP 70 site and also ligand binding. The VDR can be represented in the Bar diagrams and could also be seen through the X-ray structures of the ligand-binding domains (LBDs).

This encompasses three main domains of the extra cellular domain, plasma membrane domain, cytoplasmic or the intracellular domain. These are the key to the generation of the second messengers and the transudation of the hormones.

Vitamin D receptors forms a complex with another intracellular receptor, the retnoid-X receptor and that heterodimer, is what binds to the DNA. This activates transcription, this suppresses vitamin D. It binds with several forms of calciferol .The hormone-receptor complex binds to the promoter regions of the responsible genes and inhibits the transcription. This causes the exchange in the phenotype of the cell. Thus the mechanism regulated the gene action on he target cells.

When the hormone binds to the receptor the series of events occur as follows: -Receptor activation-this is the conformational change in the receptor due to the hormone. Binding of the activated receptor to the hormones response elements and the last event is that of the transcription of the genes to which this receptor is bound. The sequential arrangement of this hormone remains to be GGTACANNNTGTTCT.
